Despite early detection and improved therapeutic options, prostate cancer (PCa) is still the second-leading cause for cancer related deaths of men in the western society. Major problem in PCa treatment is the development of androgen resistance during androgen depletion therapy, often leading to castration-resistant prostate cancer (CRPC). Within this project, we are analyzing different cell line models with genetically modified RAI2 or CtBP protein expression and focus on the question how RAI2-controlled CtBP activity contributes to nucleosome packing and genomic integrity. More specifically, we aim to decipher the transcriptional network of CtBPs in PCa by the combination of different genome wide approaches. In parallel, we are validating in clinical samples whether measuring RAI2 gene expression might represent a new biomarker for the detection of circulating tumor cells, indicating progression to castration resistant disease.
